Brimonidine is thought to reduce the pressure in the eyes by increasing outflow of the fluid from the eyes and reducing formation of fluids there. Timolol, too, acts by reducing formation of a fluid called aqueous humour in the eyes to reduce pressure.
Timolol and its metabolites are mainly found excreted in the urine. Timolol half-life was measured at 2.9 ± 0.3 h hours in a clinical study of healthy volunteers. One pharmacokinetic study in healthy volunteers measured the total plasma clearance of timolol to be 557 ± 61 ml/min,Pharmacokinetics: After ocular administration of either a 0.1% or 0.2% solution, plasma concentrations peaked within 0.5 to 2.5 hours and declined with a systemic half-life of approximately 2 hours. In humans, systemic metabolism of brimonidine is extensive. It is metabolized primarily by the liver.
