Acugesic T 500 mg/37.5 mg Tablet can be taken with or without food. The dose and duration will depend on what you are taking it for and how well it helps your symptoms. Mechanism Of Action

Acugesic T 500 mg/37.5 mg Tablet is a combination of two medicines: Paracetamol/Acetaminophen and Tramadol. Paracetamol/Acetaminophen is an analgesic (pain reliever) which works by blocking the release of certain chemical messengers that cause pain.

Pharmacokinets

Paracetamol is readily absorbed from the gastrointestinal tract with peak plasma concentrations occurring about 30 minutes to 2 hours after ingestion. It is metabolised in the liver (90-95%) and excreted in the urine mainly as the glucuronide and sulphate conjugates. Less than 5% is excreted as unchanged paracetamol.

Tramadol is rapidly distributed in the body; plasma protein binding is about 20%. Tramadol is mainly metabolised by O- and N-demethylation and by conjugation reactions forming glucuronides and sulfates. Tramadol and its metabolites are mainly excreted via the kidneys. The mean elimination half-life is about 6 hours.

Interactions

Otherwise, no serious adverse drug interactions with therapeutic doses of paracetamol have been confirmed in humans. Because the absorption of paracetamol is so dependent on gastric emptying, other drugs that alter gastric emptying can change its pharmacokinetics; but this would not cause serious adverse effects.
