Basic Information

Mechanism Of Action

Amirid Tablet is a combination of three medicines: Diloxanide, tinidazole and simethicone. Diloxanide is a luminal amoebicide which acts inside the intestines to kill the infection-causing microorganisms (amoeba). Diloxanide is combined with tinidazole in case of severe intestinal infections.

Pharmacokinets

Human pharmacokinetic data are limited. Animal data show that diloxanide furoate is rapidly absorbed from the intestine. The furoate is hydrolyzed in the gut, leaving high intraluminal concentrations of free diloxanide. About 75% is excreted via the kidney within 48 h, mostly as a glucuronide.

Tinidazole is excreted by the liver and the kidneys. Tinidazole is excreted in the urine mainly as unchanged drug (approximately 20-25% of the administered dose). Approximately 12% of the drug is excreted in the feces. The elimination half-life is 13.2±1.4 hours and the plasma half-life is 12 to 14 hours.

Simethicone and cellulose are both excreted unchanged in the feces. Simethicone is not known to interfere with gastric secretion or nutrient absorption; however, this medicine is usually taken after at least 4 hours of fasting. SonoRx may color the stool orange until elimination is complete, typically 24 to 48 hours.
