All penicillins inhibit the biosynthesis of the bacterial cell wall. By binding to specific penicillin-binding proteins (PBPs) located inside the bacterial cell wall, dicloxacillin inhibits the third and last stage of bacterial cell wall synthesis.
Pharmacokinetics. Ampicillin is well-absorbed from the GI tract (though food reduces its absorption), and reaches peak concentrations in one to two hours. The bioavailability is around 62% for parenteral routes. Unlike other penicillins, which usually bind 60–90% to plasma proteins, ampicillin binds to only 15–20%. Dicloxacillin was excreted in its unchanged form via the kidney, with no tendency of accumulation, and varied from 38.65% to 50.10%. No appreciable accumulation of drug occurred with multiple oral doses of dicloxacillin. No serious adverse events were reported. Adverse events were generally mild.

DRUG-DRUG INTERACTIONS: AMPICILLIN+DICLOXACILLIN may have interaction with antibiotics (chloramphenicol, doxycycline), anticoagulants (warfarin), pain killer (aspirin), vaccines (live cholera vaccine), Anti-hypertensives (atenolol), anti-cancer (methotrexate).
