Basic Information

Mechanism Of Action

Ondansetron is a medication prescribed for the treatment and prevention of nausea and vomiting. It's a serotonin antagonist, meaning its mechanism of action is blocking the serotonin receptors in the CTZ.

Ranitidine is a competitive inhibitor of histamine H2-receptors. The reversible inhibition of H2-receptors in gastric parietal cells results in a reduction in both gastric acid volume and concentration.

Pharmacokinets

Ondansetron is completely and rapidly absorbed from the gastrointestinal tract after oral administration, and does not accumulate with repeated oral administration. Owing to hepatic first-pass metabolism, its bioavailability is only about 60% compared with ondansetron administered by infusion over 15 minutes.

The pharmacokinetics of ranitidine has been described previously[9-11]. After oral administration 50% of ranitidine is absorbed, resulting in the peak serum concentration of 2 ± 3 h after dosing, followed by elimination with a half-life of 2.5 ± 3 h.
