Pharmacokinetics. Methylcobalamin can be administered orally, parenterally and intranasal. Methylcobalamin binds with an intrinsic factor and form a complex which is absorbed in distal ileum. Its half life is 6 days,Thiamine requirements are related to energy metabolism; specifically, 0.33 mg of thiamine are required for every 4400 kJ of energy. For adults the DRI of thiamine is between 1.1 and 1.4 mg per day. The primary active form of the vitamin, thiamine diphosphate (ThDP), is also known as thiamine pyrophosphate (TPP),Pyridoxine is used medically for the treatment of vitamin B6 deficiency and for the prophylaxis of isoniazid-induced peripheral neuropathy (due to Isoniazid's mechanism of action which competitively inhibits the action of pyridoxine in the above-mentioned metabolic functions),The pharmacokinetics of nicotinamide were studied in four human volunteers after oral doses of 1–6 g. Plasma concentrations and clearance rates of the vitamin were found to be dose-dependent, with a half-life of ∼7–9 h for the two highest doses administered (4 and 6 g), ∼4 h with 2 g and ∼1.5 h with a 1-g dose,
