A single oral dose of 40 to 120 mg of gliclazide results in a Cmax of 2.2 to 8.0 µg/ml within 2 to 8 hours. Tmax and cmax are increased after repeated gliclazide administration. Steady state concentration is achieved after 2 days administration of 40 to 120 mg of gliclazide.Metformin has an oral bioavailability of 50–60% under fasting conditions, and is absorbed slowly. Peak plasma concentrations (Cmax) are reached within 1–3 hours of taking immediate-release metformin and 4–8 hours with extended-release formulations.
