The bioavailability of doxylamine is 24.7% for oral administration and 70.8% for intranasal administration. The Tmax of doxylamine is 1.5 to 2.5 hours. Its elimination half-life is 10 to 12 hours (range 7 to 13 hours).Pyridoxine is rapidly metabolized in erythrocytes. 4-Pyridoxic acid and pyridoxine are secreted in renal tubulus, whereas pyridoxal is reabsorbed. Metabolism of vitamin B6 is altered in premature infants. Vitamin B6 supplements are of multipurpose use today. The pharmacokinetic profile of plasma FA after oral administration indicated an absorption rate-limited elimination, i.e., so called 'flip-flop' phenomenon. The bioavailability of FA was quite low after a high oral dose (F = 0.01), in contrast to a high value after intramuscular administration (F = 0.95).
