The DA2-receptor antagonist domperidone antagonizes the inhibitory effect of dopamine, resulting in stimulation of gastric muscle contraction. This provides a mechanism for the gastrokinetic effect of domperidone
Cinnarizine inhibits smooth muscle cell contraction in the vasculature by blocking L-and T-type voltage-gated calcium channel. It is also known to bind to histamine H1 receptors, muscarinic (acetyl choline) receptors and dopamine D2 receptors
The absorption of domperidone was very rapid, with mean peak plasma concentration (Cmax) values of 18.8, 15.0, and 20.7 ng/mL attained at 0.9, 1.2, and 0.6 hours after the administration of base tablet, maleate tablet, and solution, respectively. The mean elimination half-life (t1/2) ranged from 12.6 to 16.0 hours
Cinnarizine is most commonly taken orally, in tablet form, with frequency and amount of dosage varying depending on the reason for taking the medication. Once ingested, the substance is absorbed quite rapidly and reaches a peak plasma concentration in 1–3 hours post-administration.
Most side effects do not require any medical attention and disappear as your body adjusts to the medicine. Consult your doctor if they persist or if you’re worried about them
