The basis for these investigations is the variety of potential mechanisms of action of nicotinamide and zinc, including an anti-inflammatory effect via inhibition of leukocyte chemotaxis, lysosomal enzyme release, lymphocytic transformation, mast cell degranulation, bacteriostatic effect against Propionibacterium acnes
Clindamycin works primarily by binding to the 50s ribosomal subunit of bacteria. This agent disrupts protein synthesis by interfering with the transpeptidation reaction, which thereby inhibits early chain elongation
The pharmacokinetics of nicotinamide were studied in four human volunteers after oral doses of 1–6 g. Plasma concentrations and clearance rates of the vitamin were found to be dose-dependent, with a half-life of ∼7–9 h for the two highest doses administered (4 and 6 g), ∼4 h with 2 g and ∼1.5 h with a 1-g dose
Clindamycin is absorbed well orally and can be given parenterally. Clindamycin diffuses well into body fluids except cerebrospinal fluid; it is concentrated in phagocytes. Most of the drug is metabolized; metabolites are excreted in bile and urine
