Trifluoperazine blocks postsynaptic mesolimbic dopaminergic D1 and D2 receptors in the brain; depresses the release of hypothalamic and hypophyseal hormones and is believed to depress the reticular activating system thus affecting basal metabolism, body temperature, wakefulness, vasomotor tone, and emesis.
Anti-anxiety action of chlordiazepoxide is believed to result from stimulation of GABA receptors in the ascending reticular activating system, since GABA in inhibitory receptor stimulation increases inhibition and blocks both cortical and limbic arousal following stimulation of the brainstem reticular formation
Chlordiazepoxide is a long-acting benzodiazepine drug. The half-life of Chlordiazepoxide is 5 – 30 hours but has an active benzodiazepine metabolite (desmethyldiazepam), which has a half-life of 36 – 200 hours
The pharmacokinetics for trifluoperazine are typical of phenothiazines such as chlorpromazine. It is readily absorbed from the gastrointestinal tract with peak plasma levels being reached from 1.5 to 6.0 hours after ingestion. A high interindividual variation in bioavailability has been consistently reported
