Mechanism Of Action

Cofmac Syrup is a combination of four medicines: Ammonium chloride, chlorpheniramine, dextromethorphan and guaifenesin which relieves cough. Ammonium chloride and guaifenesin are expectorants which decrease the stickiness of mucus (phlegm) and help in its removal from the airways.

Pharmacokinets

Ammonium chloride increases acidity by increasing the amount of hydrogen ion concentrations. Ammonium chloride can be used as an expectorant due to its irritative action on the bronchial mucosa. This effect causes the production of respiratory tract fluid which in order facilitates the effective cough.Chlorpheniramine has a serum half-life of approximately 20 hours in adults, and elimination from the body is primarily by metabolism to monodesmethyl and didesmethyl compounds. The half-life is increased in the presence of renal dysfunction and decreased in children.Following oral administration, dextromethorphan is rapidly absorbed from the gastrointestinal tract, where it enters the bloodstream and crosses the blood–brain barrier.Guaifenesin is administered orally. The plasma half-life is approximately 1 hour. Guiafenesin is rapidly hydrolyzed (60% within seven hours) and then excreted in the urine, with beta-(2-methoxyphenoxy)-lactic acid as its major urinary metabolite. No unchanged drug was detected in the urine following administration.

Effects Of Missed Dosage
