Colibest Tablet can be taken with or without food, but better to take it at the same time every day for better results. The dose and how often you take it depend on what you are taking it for. Your doctor will decide how much you need to improve your symptoms. You should take this medicine for as long as it is prescribed for you.The most common side effects of this medicine include nausea, vomiting, and dryness in the mouth. If these bother you or appear serious, let your doctor know. There may be ways of reducing or preventing them. It may also make you feel dizzy due to low blood pressure, if that happens take precautions while driving.
COLIBEST relieves abdominal pain, where Drotaverine provides relief from muscle spasms by inhibiting multisynaptic reflex on the brain and spinal cord centers, and Nimesulide provides relief from abdominal pain by blocking the release of certain chemical messengers that are responsible to cause pain and inflammation in the affected areas.
Drotaverine is mainly eliminated via hepatic metabolism. About 67% of the drug is found in feces and 20% of the drug was eliminated with urine. Following oral administration of a single 80 mg dose, the mean half-life was 9.11 ± 1.29 hours. Following an intravenous dose of 80 mg, the mean half-life 9.33 ± 1.02 hours.Nimesulide is rapidly distributed and has an apparent volume of distribution ranging between 0.18 and 0.39 L/kg. It is extensively bound to albumin; the unbound fraction in plasma was 1%. The unbound fraction increased to 2 and 4% in patients with renal or hepatic insufficiency.

Elevated levels of fat in the blood
Fits
Gallbladder disease
Hemolytic uremic syndrome
Colibest Tablet is unsafe to use during pregnancy as there is definite evidence of risk to the developing baby. However, the doctor may rarely prescribe it in some life-threatening situations if the benefits are more than the potential risks. Please consult your doctor.
It is unsafe to consume alcohol with Colibest Tablet.

If you use other drugs or over the counter products at the same time, the effects of Colibest Tablet may change. This may increase your risk for side-effects or cause your drug not to work properly. Tell your doctor about all the drugs, vitamins, and herbal supplements you are using, so that you doctor can help you prevent or manage drug interactions. Colibest Tablet may interact with the following drugs and products:
Analgesics
Antimuscarinics
Benzodiazepines
Cyclosporine
Digitalis glycosides
Levodopa
Lithium
Methotrexate
Phenytoin
Do not chew, crush or break it. Colibest Tablet is to be taken empty stomach.
Store below 30°C
If you miss a dose of Colibest Tablet, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose and go back to your regular schedule. Do not double the dose.

Drotaverine
Drotaverine helps relieve pain due to smooth muscle spasms such as menstrual pain, pain due to kidney stone, and colicky pain.
Take it as per dose and duration suggested by your doctor.
It may cause a drop in your blood pressure. Monitor your blood pressure regularly and inform your doctor if you have a history of low blood pressure.
Use caution while driving or doing anything that requires concentration as Drotaverine may cause dizziness.
Inform your doctor if you have severe kidney, liver, or heart disease.
Nimesulide
Nimesulide is used to relieve pain, inflammation, and fever.
Take it with food or milk to prevent upset stomach.
Take it as per the dose and duration prescribed by your doctor. Long term use may lead to serious complications such as stomach bleeding and kidney problems.
Inform your doctor if you have a history of heart disease or stroke.
Avoid consuming alcohol while taking Nimesulide as it can increase your risk of stomach problems.
Your doctor may regularly monitor your kidney function, liver function and levels of blood components if you are taking this medicine for long-term treatment.
