Colkoff Capsule is taken with or without food in a dose and duration as advised by the doctor. The dose you are given will depend on your condition and how you respond to the medicine. You should keep taking this medicine for as long as your doctor recommends. If you stop treatment too early your symptoms may come back and your condition may worsen. Let your healthcare team know about all other medications you are taking as some may affect, or be affected by this medicine.The most common side effects are nausea, upset stomach, insomnia, palpitations, and high blood pressure. Most of these are temporary and usually resolve with time. Contact your doctor straight away if you are at all concerned about any of these side effects. This medicine can also cause dizziness and sleepiness, so do not drive or do anything that requires mental focus until you know how this medicine affects you. Avoid drinking alcohol while taking this medicine as it can worsen your sleepiness.
Colkoff Capsule is a combination of three medicines: Phenylpropanolamine, Chlorpheniramine Maleate and Dextromethorphan Hydrobromide which relieve dry cough. Phenylpropanolamine is a decongestant which narrows the small blood vessels providing relief from congestion or stuffiness in the nose. Chlorpheniramine is an antiallergic which relieves allergy symptoms like runny nose, watery eyes and sneezing. Dextromethorphan is a cough suppressant that relieves cough by reducing the activity of cough centre in the brain.
The drug was shown to be rapidly absorbed with a mean tmaxof 1.47 ± 0.49 h and a mean elimination half-life of 4.0 ± 0.5 h. Phenylpropanolamine is predominantly excreted via the kidney with a mean renal clearance of 0.646 ± 0.089 liter/kg/h and 90.2 ± 1.7% excreted unchanged in the urine.Chlorpheniramine has a serum half-life of approximately 20 hours in adults, and elimination from the body is primarily by metabolism to monodesmethyl and didesmethyl compounds. The half-life is increased in the presence of renal dysfunction and decreased in children.Dextromethorphan is rapidly absorbed from the gastrointestinal tract and converted into the active metabolite dextrorphan in the liver by the cytochrome P450 enzyme CYP2D6. The average dose necessary for effective antitussive therapy is between 10 and 45 mg, depending on the individual.
Most side effects do not require any medical attention and disappear as your body adjusts to the medicine. Consult your doctor if they persist or if you’re worried about them

Hypersensitivity to Colkoff Capsule is a contraindication. In addition, Colkoff Capsule should not be used if you have the following conditions:
Blockage of urinary bladder
Cannot empty bladder
Chronic difficulty having a bowel movement
Chronic obstructive lung disease
Closed angle glaucoma
Concomitant use with a monoamine oxidase inhibitor
Enlarged prostate
Glaucoma
High blood pressure
Hypertension
Colkoff Capsule may be unsafe to use during pregnancy. Although there are limited studies in humans, animal studies have shown harmful effects on the developing baby. Your doctor will weigh the benefits and any potential risks before prescribing it to you. Please consult your doctor.
Caution is advised when consuming alcohol with Colkoff Capsule. Please consult your doctor.

If you use other drugs or over the counter products at the same time, the effects of Colkoff Capsule may change. This may increase your risk for side-effects or cause your drug not to work properly. Tell your doctor about all the drugs, vitamins, and herbal supplements you are using, so that you doctor can help you prevent or manage drug interactions. Colkoff Capsule may interact with the following drugs and products:
Acebutolol
Alcohol
Amiodarone
Antidepressants
Atropine
Bromocriptine
Cough and cold medicines
Debrisoquine
Diazepam
Doxepin
Colkoff Capsule may be taken with or without food, but it is better to take it at a fixed time.
Store below 30°C
If you miss a dose of Colkoff Capsule, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose and go back to your regular schedule. Do not double the dose.
Consult a doctor before taking any medicines.
Colkoff Capsule is prescribed to get relief from a dry cough.
Take exactly as directed by your doctor or on the label. Do not increase the dosage or take for longer than is recommended.
Consult your doctor if you do not see any improvement and have a cough for more than 7 days.
Let your doctor know if you are taking any anti-depressants.
Inform your doctor if you have had a cough for a long time or if you have asthma or suffer from serious asthma attacks, or if your cough is accompanied by a fever, rash, or persistent headache.
