Colotecan 40mg Injection is given as an IV infusion into vein by a qualified medical professional. Your doctor will decide what dose is necessary and how often you need to take it. Taking it in the wrong way or taking too much can cause very serious side effects. It may take several weeks or months for you to see or feel the benefits but do not stop taking it unless your doctor tells you to.The most common side effects of this medicine include fatigue, nausea, vomiting, weakness, and hair loss. This medicine may reduce the number of blood cells (decrease red blood and white blood cells) in your blood, thereby, increasing the susceptibility to infections. Inform your doctor if you notice fever, sore throat, and continuous episodes of diarrhea. Regular blood tests are required to check your blood cells along with heart, liver, and blood uric acid levels.

Mechanism Of Action

Colotecan 40mg Injection is an anti-cancer medication. It works by suppressing the activity of an enzyme (topoisomerase I) involved in DNA multiplication of the cancer cells. This slows the growth of cancer cells and eventually kills them.

Pharmacokinets

The clinical pharmacokinetics of irinotecan (CPT11) can be described by a 2 or 3 compartment model, a mean terminal half-life of 12 hours, a volume of distribution at steady state of 168 l/m2 and a total body clearance of 15 l/m2/h. Irinotecan is 65% bound to plasma proteins.

Indications

Irinotecan is indicated for patients with metastatic carcinoma of the colon or rectum whose disease has recurred or progressed following initial fluorouracil-based therapy.

Interactions

Some medicines can affect the way Colotecan Injection works or this medicine itself can reduce the effectiveness of other medicines taken at the same time.

Tell your doctor about all medicines, supplements, or herbals you are currently taking or might take to avoid any possible interaction.

Particular care should be taken if you are taking medicines used to treat fits/seizures (carbamazepine, phenobarbital, phenytoin, and fosphenytoin), medicines used to treat fungal infections (ketoconazole, itraconazole, voriconazole and posaconazole), medicines used to treat bacterial infections (clarithromycin, erythromycin, and telithromycin), medicines used to treat tuberculosis (rifampicin and rifabutin), St. John's Wort (a herbal medicine), live attenuated vaccines, medicines used to treat HIV (indinavir, atazanavir, and others), medicines used to suppress your body’s immune system to prevent transplant rejection (cyclosporine and tacrolimus), medicines used to treat cancer (regorafenib, crizotinib, idelalisib, cetuximab, and apalutamide), vitamin K antagonists (blood thinners like warfarin), medicines used to relax muscles used during general anaesthesia and surgery (suxamethonium), 5-fluorouracil/folinic acid, and bevacizumab (a blood vessel growth inhibitor).

Avoid taking laxatives and stool softeners while taking Colotecan Injection.

Inform your doctor if you notice passage of blood in stools or experience dizziness or faintness, continuous episodes of nausea, vomiting, diarrhea or fever.
