Basic Information

Mechanism Of Action

Menthol is a selective activator of transient receptor potential melastatin-8 (TRPM8) channels and is also a vasoactive compound. As a topical agent, it acts as a counter-irritant by imparting a cooling effect and by initially stimulating nociceptors and then desensitizing them.

This medication works by blocking a certain natural substance (histamine) that your body makes during an allergic reaction. By blocking another natural substance made by your body (acetylcholine), it helps dry up some body fluids to relieve symptoms such as watery eyes and runny nose

Pharmacokinets

In conclusion, the pharmacokinetics of codeine were similar to other opioids in dogs with a short half-life, rapid clearance, large volume of distribution, and poor oral bioavailability. High concentrations of codeine-6-glucuronide were detected after IV and oral administration.

Some studies show that menthol acts as GABAA receptor positive allosteric modulator and increases Gabaergic transmission in PAG neurons. Menthol also shares anaesthetic properties similar to propofol, by modulating the same sites of the GABAA receptor.
