Basic Information

Mechanism Of Action

Methylcobalamin is an active form of vitamin B12, and a coenzyme of methionine synthase, which is required for DNA and protein methylation. In addition, methylcobalamin facilitates neurite outgrowth and inhibits neural apoptosis through the Erk1/2 and Akt signaling pathways.

Vitamin B6 plays an important role in the body. It is needed to maintain the health of nerves, skin, and red blood cells. Pyridoxine has been used to prevent or treat a certain nerve disorder (peripheral neuropathy) caused by certain medications

Within a complex metabolic system niacinamide controls the NFκB-mediated transcription of signalling molecules by inhibiting the nuclear poly (ADP-ribose) polymerase-1 (PARP-1). Niacinamide is a well-tolerated and safe substance often used in cosmetics.

Pharmacokinets

Methylcobalamin can be administered orally, parenterally and intranasal. Methylcobalamin binds with an intrinsic factor and form a complex which is absorbed in distal ileum. Its half life is 6 days.

There is no indication of circadian rhythm in vitamin B6-levels in blood plasma. Pharmacokinetics of vitamin B6 differ fundamentally, depending from the route of administration. The rate constants of elimination of B6-metabolites indicate a tendency of accumulation. Pyridoxine is rapidly metabolized in erythrocytes

Nicotinamide, the amide form of vitamin B3 (niacin), has long been associated with neuronal development, survival, and function in the central nervous system (CNS), being implicated in both neuronal death and neuroprotection.

Side Effects

No common side effects seen

Indications

MeCbl is now prescribed for the treatment of vitamin B12 deficiency and is slowly replacing the traditional B12 formulations like cyanocobalamin and hydroxocobalamin. MeCbl is essential for the normal functioning of the brain and nervous system and the formation of blood

Vitamin B6 is indicated in cases of its deficiency, which may be due to poor renal function, autoimmune diseases, increased alcohol intake, or isoniazid, cycloserine, valproic acid, phenytoin, carbamazepine, primidone, hydralazine, and theophylline therapy

People use niacinamide to prevent vitamin B3 deficiency and related conditions such as pellagra. It is also used for acne, diabetes, cancer, osteoarthritis, aging skin, skin discoloration, and many other conditions, but there is no good scientific evidence to support most of these uses.
