Delpodol P 325mg/37.5mg Tablet can be taken with or without food. The dose and duration will depend on what you are taking it for and how well it helps your symptoms. You should keep taking the medicine even if you feel better until the doctor says it is alright to stop using it.

Mechanism Of Action

Paracetamol has a central analgesic effect that is mediated through activation of descending serotonergic pathways. Debate exists about its primary site of action, which may be inhibition of prostaglandin (PG) synthesis or through an active metabolite influencing cannabinoid receptors

Analgesics can inhibit the sensation of pain by inhibiting transmission of non-nociceptive impulses along primary afferents or by altering the perception of pain

Tramadol modulates the descending pain pathways within the central nervous system through the binding of parent and M1 metabolite to μ-opioid receptors and the weak inhibition of the reuptake of norepinephrine and serotonin

Pharmacokinets

Tramadol is rapidly distributed in the body; plasma protein binding is about 20%. Tramadol is mainly metabolised by O- and N-demethylation and by conjugation reactions forming glucuronides and sulfates. Tramadol and its metabolites are mainly excreted via the kidneys. The mean elimination half-life is about 6 hours

Paracetamol is readily absorbed from the gastrointestinal tract with peak plasma concentrations occurring about 30 minutes to 2 hours after ingestion. It is metabolised in the liver (90-95%) and excreted in the urine mainly as the glucuronide and sulphate conjugates. Less than 5% is excreted as unchanged paracetamol.

Acetaminophen has a high oral bioavailability (88%), it is well absorbed and reaches the peak blood concentrations within 90 minutes after ingestion.

APAP is not widely bound to plasma proteins, and has a plasma half-life of 1.5–2.5 hours at the recommended doses

It is unsafe to consume alcohol with Delpodol P 325mg/37.5mg Tablet.


Breast Feeding Related Information

Delpodol P 325mg/37.5mg Tablet is probably safe to use during breastfeeding. Limited human data suggests that the drug does not represent any significant risk to the baby.

Consult a doctor immediately if the baby shows signs of increased sleepiness (more than usual), difficulty breastfeeding, breathing problems, or limpness.


Indications

Tramadol/Paracetamol tablets are indicated for the symptomatic treatment of moderate to severe pain. The use of Tramadol/Paracetamol should be restricted to patients whose moderate to severe pain is considered to require a combination of tramadol and paracetamol


Interactions

Tramadol should not be used in combination with MAO inhibitors or serotonin-precursors (such as L-tryptophan, oxitriptan) and should be used with caution in combination with other serotonergic drugs

Despite its frequent coadministration with other drugs and widespread use, very few clinically significant drug interactions have been documented with paracetamol. Drugs that alter gastric emptying can alter paracetamol pharmacokinetics, but this does not result in toxicity in therapeutic use.

Drug interactions of Tylenol include carbamazepine, isoniazid, rifampin, alcohol, cholestyramine, and warfarin. Tylenol is often used during pregnancy for short-term treatment of fever and minor pain during pregnancy.

Effects Of Missed Dosage

If you miss a dose of Delpodol P 325mg/37.5mg Tablet, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose and go back to your regular schedule. Do not double the dose.
