Depnil Forte 5 mg/25 mg Tablet can be taken with or without food. However, it is advised to take it at the same time each day as this helps to maintain a consistent level of medicine in the body. Take this medicine in the dose and duration advised by your doctor as it has habit-forming potential. If you miss a dose of this medicine, take it as soon as you remember. Finish the full course of treatment even if you feel better. It is important not to stop taking this medication suddenly without talking to your doctor as this can cause withdrawal effects like depression and difficulty sleeping.

Mechanism Of Action

Depnil Forte 5 mg/25 mg Tablet is a combination of two medicines: Diazepam and imipramine which treat depression. Diazepam is a benzodiazepine (BZD) which works by increasing the action of GABA, a chemical messenger which suppresses the abnormal and excessive activity of the nerve cells in the brain. Imipramine is a tricyclic antidepressant which increases the levels of chemical messengers in the brain that help in regulating the mood and treat depression.


Pharmacokinets

Diazepam and its metabolites are excreted mainly in the urine, predominantly as their glucuronide conjugates. The clearance of diazepam is 20 to 30 mL/min in young adults. Diazepam accumulates upon multiple dosing and there is some evidence that the terminal elimination half-life is slightly prolonged.The mean elimination half-life for imipramine was significantly decreased in alcoholics (8.7 vs. 19.9 hours after intravenous infusion and 10.9 vs. 19.6 hours after oral administration; P less than 0.05). The mean elimination half-life for desipramine was decreased in alcoholics after intravenous infusion

Contra Indications

DIAZEPAM+IMIPRAMINE should not be used in patients with severe liver disease, heart disease, mania (exaggerated behavior or hyperactivity), porphyria (an inherited blood disorder), difficulty passing urine, glaucoma (increased pressure inside the eye), severe breathing problems, phobia (a fear of particular object), myasthenia gravis (a condition in which muscles weaken), and sleep apnoea (a sleep disorder).

Pregnancy Related Information

Depnil Forte 5 mg/25 mg Tablet is unsafe to use during pregnancy as there is definite evidence of risk to the developing baby. However, the doctor may rarely prescribe it in some life-threatening situations if the benefits are more than the potential risks. Please consult your doctor.


Product And Alcohol Interaction

It is unsafe to consume alcohol with Depnil Forte 5 mg/25 mg Tablet.


Breast Feeding Related Information

Depnil Forte 5 mg/25 mg Tablet is probably unsafe to use during breastfeeding. Limited human data suggests that the drug may pass into the breastmilk and harm the baby.

Interactions

DIAZEPAM+IMIPRAMINE may interact with an opioid pain killer (buprenorphine), a medicine used to treat sleep problems (sodium oxybate), other antidepressants (fluvoxamine, fluoxetine, phenelzine, moclobemide, and Hypericum perforatum), antipsychotics (clozapine), medicines used to treat allergies, general anesthetics, medicines used to induce sleep (alprazolam), antibiotics (erythromycin and rifampicin), medicines used to relax muscles (suxamethonium and tubocurarin), strong pain killer (morphine), medicines used to treat fits or mental disorders (phenobarbital), medicines used to treat high blood pressure, antacids (cimetidine, omeprazole, and esomeprazole), a medicine used to treat alcohol addiction (disulfiram), medicines used to treat fits (phenytoin, carbamazepine, and sodium valproate), medicine used to treat asthma (theophylline), antivirals (atazanavir, ritonavir, efavirenz, indinavir, and zidovudine, etc.), antitubercular (isoniazid), oral contraceptives, steroid medicines, anesthetic (ketamine), medicines to treat nausea and vomiting (nabilone), and medicines used to treat heart conditions (diltiazem, verapamil, and quinidine, etc.).

Effects Of Missed Dosage

If you miss a dose of Depnil Forte 5 mg/25 mg Tablet, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose and go back to your regular schedule. Do not double the dose.

If you have hypertension, monitor your blood pressure regularly while taking Depnil Forte 5 mg/25 mg Tablet.
