Clotrimazole exerts its action primarily by damaging the permeability barrier in the fungal cytoplasmic membrane. Clotrimazole thereby inhibits the biosynthesis of ergosterol in a concentration-dependent manner by inhibiting the demethylation of 14 alpha lanosterol
Beclomethasone dipropionate works by attenuating the inflammatory responses associated with asthma, allergic rhinitis, nasal polyps, and corticosteroid-responsive dermatoses. It suppresses the actions of inflammatory cells, such as mast cells, eosinophils, basophils, lymphocytes, macrophages, and neutrophils
Neomycin is bactericidal in action. Similar to other aminoglycosides, it inhibits bacterial protein synthesis through irreversible binding to the 30 S ribosomal subunit of susceptible bacteria
After vaginal treatment with clotrimazole, the small fraction absorbed into the systemic circulation--between 3% and 10% of the dose--is subjected to metabolism and excretion as after oral or intravenous administration. The vaginal availability of clotrimazole is largely dependent on the formulation applied
Rapidly absorbed from pulmonary, nasal, and GI tissue. Undergoes extensive first-pass metabolism in the liver. Protein binding: 87%. Primarily eliminated in feces.
There is limited information on the metabolism of neomycin, as there is limited systemic absorption following drug administration. Metabolism is deemed to be negligible. The small absorbed fraction of neomycin is excreted by the kidney. The unabsorbed portion of the drug is excreted unchanged in the feces.
