Diclofenac works by blocking the release of chemical messengers, which are responsible for causing pain and inflammation. Tramadol, on the other hand, blocks the transmission of pain signals to the brain. As a result, pain perception is lowered, and hence the quality of life is improved.
Diclofenac binds extensively to plasma albumin. The area under the plasma concentration-time curve (AUC) of diclofenac is proportional to the dose for oral doses between 25 to 150 mg. Substantial concentrations of drug are attained in synovial fluid, which is the proposed site of action for NSAIDs.
Tramadol is rapidly distributed in the body; plasma protein binding is about 20%. Tramadol is mainly metabolised by O- and N-demethylation and by conjugation reactions forming glucuronides and sulfates. Tramadol and its metabolites are mainly excreted via the kidneys. The mean elimination half-life is about 6 hours.
