Basic Information

Mechanism Of Action

Lupicet Cold Tablet is a combination of three medicines: Paracetamol / Acetaminophen, Phenylpropanolamine and Cetirizine which relieve common cold symptoms. Paracetamol / Acetaminophen is a analgesic (pain reliever) and anti-pyretic (fever reducer) which works by blocking the release of certain chemical messengers that causes fever and pain. Phenylpropanolamine is a sympathomimetic agonist. It narrows blood vessels in the nose to relieve congestion. Cetirizine is an antiallergic which relieves allergy symptoms like runny nose, watery eyes and sneezing.

Pharmacokinets

It distributes rapidly and evenly throughout most tissues and fluids and has a volume of distribution of approximately 0.9L/kg. 10 to 20% of the drug is bound to red blood cells. Paracetamol is extensively metabolised (predominantly in the liver), the major metabolites being the sulphate and glucuronide conjugates.Phenylpropanolamine is a sympathomimetic agent that acts as a nonselective adrenergic receptor agonist and norepinephrine reuptake inhibitor. It has been used as a decongestant and appetite suppressant. Currently, it is withdrawn from the market in Canada and the United States due to the risk for hemorrahgic strokes. The Tmax of cetirizine is approximately 1.0 hour regardless of formulation. The pharmacokinetics of cetirizine have been found to increase linearly with dose across a range of 5 to 60 mg. Its Cmax following a single dose has been found to be 257 ng/mL for 10 mg and 580 ng/mL for 20 mg.
