Nildep 10mg Tablet is normally taken before bedtime because it can make you feel sleepy. It can be taken with or without food. The dose and how often you need to take it will be decided by your doctor so that you get the right amount to control your symptoms. The initial dose should be increased with great caution under close supervision. This medicine may not work straight away and it could be several weeks before you get the full effects.
How long you need to use it will depend on what you are taking it for, but it is important to continue taking this medication even if you feel well. Do not stop taking the medicine suddenly without talking to your doctor. Stopping the medicine suddenly can worsen your conditions or cause other symptoms. Your dose may be modified or gradually decreased before stopping the medication.
The most common side effects of this medicine include nausea, constipation, sweating, dry mouth, sleepiness, tremor, dizziness, headache, speech disorder, accommodation disorder, palpitations, increased heart rate, orthostatic hypotension, and nasal congestion. It may also make you aggressive or confused and can lead to weight gain in some people.
This medicine is not addictive, but you can experience additional side effects (withdrawal symptoms) if you stop taking it suddenly. Most of the common side effects tend to be mild. Your doctor may be able to suggest ways of preventing or reducing side effects if they bother you or do not go away. Serious side effects associated with this medicine are rare.
Nildep 10mg Tablet contains Amitriptyline, a tricyclic antidepressant. It has anticholinergic and sedative properties. It prevents the reuptake, and hence the inactivation of neurotransmitters called noradrenaline and serotonin, that are present at the nerve terminals in our brain. Reuptake prevention of these monoamine neurotransmitters potentiate their action in the brain. This appears to be associated with the antidepressant activity and helps in regulating the mood. It also stops the transmission of pain signals from nerves to the brain thereby relieving neuropathic pain (pain from damaged nerves).
Imipramine has a mean half life of 12 h. Its active metabolite, desipramine has a mean half life of 22.5 h 3. Imipramine has a mean clearance of 1 L/h/kg. Its active metabolite, desipramine has a mean clearance of 1.8 L/h/kg 3.
Most side effects do not require any medical attention and disappear as your body adjusts to the medicine. Consult your doctor if they persist or if you’re worried about them

Imipramine is contraindicated in patients who have shown prior hypersensitivity to it and in acute porphyria, hepatic impairment and cardiovascular disease.
Nildep 10mg Tablet may be unsafe to use during pregnancy. Although there are limited studies in humans, animal studies have shown harmful effects on the developing baby. Your doctor will weigh the benefits and any potential risks before prescribing it to you. Please consult your doctor.
It is unsafe to consume alcohol with Nildep 10mg Tablet.
Nildep 10mg Tablet is probably unsafe to use during breastfeeding. Limited human data suggests that the drug may pass into the breastmilk and harm the baby.
Imipramine may be used to treat depression and nocturnal enuresis in children Label. Unlabeled indications include chronic and neuropathic pain (including diabetic neuropathy), panic disorder, attention-deficit/hyperactivity disorder (ADHD), and post-traumatic stress disorder (PTSD) 12,11,1,13,14,15,2.
Drug-Drug Interactions: IMIPRAMINE may interact with anti-depressants (fluoxetine, fluvoxamine, duloxetine, escitalopram, sertraline), antipsychotics (aripiprazole, quetiapine), anti-convulsant (carbamazepine, phenytoin, phenobarbital, pregabalin, topiramate), benzodiazepine (diazepam, nitrazepam, oxazepam, alprazolam, clonazepam), pain killer (aspirin, tramadol, codeine, dihydrocodeine), anti-HIV (ritonavir), antacid (cimetidine, esomeprazole), blood thinner (warfarin), skeletal muscle relaxant (cyclobenzaprine), anti-anxiety (alprazolam) and hormone (levothyroxine).
Drug-Food Interactions: Do not take St. John’s wort (herbal supplement to treat depression) along with IMIPRAMINE.
Drug-Disease Interactions: Inform your doctor if you have epilepsy, low blood pressure, overactive thyroid, urination problems or enlarged prostate, glaucoma, diabetes, schizophrenia, mania, pheochromocytoma (tumour of the adrenal gland), heart, kidney or liver problems.

If you miss a dose of Nildep 10mg Tablet, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose and go back to your regular schedule. Do not double the dose.\n
Consult a doctor before taking any medicines.
It may take 4 to 6 weeks for Nildep 10mg Tablet to start working. Keep taking it as prescribed.
It may cause sleepiness. Take it at bedtime and do not drive or do anything requiring concentration until you know how it affects you.
Avoid consuming alcohol when taking Nildep 10mg Tablet as it may cause excessive sleepiness.
Monitor your weight during the treatment as this medicine can cause weight gain and increased appetite.
It may cause sudden drop in your blood pressure. Rise slowly if you have been sitting or lying down.
Talk to your doctor if you notice sudden mood changes or develop suicidal thoughts.
Pay close attention to any changes, especially sudden changes, in mood, behaviors, thoughts, or feelings. This is very important when an antidepressant medicine is started or when the dose is changed.
Take this medicine in the dose and duration as advised by your doctor. Swallow it as a whole. Do not chew, crush or break it. Nildep 10mg Tablet may be taken with or without food, but it is better to take it at a fixed time.

Certain lifestyle changes can help you get maximum benefit of Nildep 10mg Tablet and help you recover fast. Keeping active and eating a healthy diet can make a significant difference to how quickly you recover from depression. Think positively and try to discuss your thoughts with others to reduce stress. Practice yoga or take up a hobby. Ensure that you have a sound sleep to calm your mind. Stay away from smoking or alcohol consumption as these will only worsen your depression. Take your medicines as prescribed even if you start feeling better.
Q. Is there anything I need to be careful about while on therapy with Nildep 10mg Tablet?
Nildep 10mg Tablet may cause drowsiness and dizziness, especially in the beginning of the treatment. So, avoid driving or using machinery until you know how the medicine affects you. Also, it is not advisable to drink alcohol during treatment with this medicine as it might increase sleepiness.
Q. Can the use of Nildep 10mg Tablet affect my sex life?
Yes, Nildep 10mg Tablet can affect sex life in both men and women. It may cause decreased sexual desire or you may experience discomfort during intercourse. Men may experience inability to develop or maintain an erection during sexual activity and may be unable to have an orgasm. If you notice these symptoms, do not stop taking the medicine but consult your doctor.
Q. I have been taking Nildep 10mg Tablet for depression since a week and recently I have been wanting to harm myself. What should I do? Is it because of the medicine?
You should immediately consult your doctor or go to the hospital. Your suicidal tendencies may have increased because of your disease or the medicine. Generally, Nildep 10mg Tablet takes about 2 weeks to show its effect but may take longer. Therefore, when you start taking Nildep 10mg Tablet, you may have thoughts about killing or harming yourself. This is more common in people who previously had suicidal tendencies or are young adults (less than 25 years old). However, the effect of medicine is different for every individual.
