Mechanism Of Action

Sildenafil is a selective inhibitor of cGMP-specific phosphodiesterase (PDE-5). Penile erection involves relaxation of the corpus cavernosum, an event mediated by NO and cGMP. The biological actions of cGMP are terminated by phosphodiesterase enzymes and PDE-5 is the major cGMP metabolising enzyme in this tissue.

Sildenafil was rapidly cleared from the body with a plasma elimination half-life of approximately 3–4 h over the dose range of 25 and 200 mg. These characteristics represent pharmacokinetic support for the effectiveness and appropriateness of sildenafil for the as-needed treatment of patients with ED.

Indications

Interactions

If you take sildenafil for pulmonary arterial hypertension, do not take Viagra® or other PDE5 inhibitors, such as tadalafil (Cialis®) or vardenafil (Levitra®). Viagra® also contains sildenafil. If you take too much sildenafil or take it together with these medicines, the chance for side effects will be higher.

Do not stop taking the medication suddenly without talking to your doctor as that may lead to worsening of your symptoms.
